Studies on Agglutination of Lymphocytes by Korea Native Phytagglutinins

Abstract
It has been accumulated a number of research works on phytagglutinin (PHA) reported by many investigators. Recently, the utilization of PHA in the medical field is increasing, especially as a lectin for the blood grouping, as a mitogen for artificial culture of the various cells and as a determinant of the receptor specificity of various tumor cells. The purpose of the present paper is to describe PHA can applicated as a leucoagglutinin and as a reagent for lymphocyte cytotoxicity test. 2 grams of ground seed of Korean Celosia cristata L., Vicia amurensis OETTINGEN, Glycine Max. MERR. and ground stems of Fritillaria ussuriensis Max. was mixed well with 100ml of pH 7.0 phosphate buffer saline (PBS) respectively, and the extract was treated for 30 minutes in water bath adjusted 560 C, then centrifuged, the supernatant was filtered with millipore filter paper (HA type, size 0. 45gm). With the plant extracts, agglutination of human lymphocyte, chicken thymus and bursa cell, and agglutination inhibition with various sugars, and lymphocyte cytotoxicity test were carried out and the following results were observed. All of human lymphocytes, chicken thymus and bursa cells were agglutinated by extracts of Celosia cristata and Vicia amurensis, however, only chicken thymus cells among them were agglutinated by extract of Glycine Max. In the cytotoxicity test of 214 of human lymphocytes with Fritillaria ussuriensis extract, 67% was positive, which showed the extract acts as a anti-HLA lectin, and the frequency suggested that a new specificity of lymphocyte was demonstrated.
