Genetic Characteristics of DNA Fingerprints in Korean detected by pV47-2 Multilocus Probe

Abstract
DNA polymorphisms arising from variations in the number of short tandem repeat account for most of VNTRs loci existed in human genome that have known as individua1 specific and highly informative genetic markers. Hybridization probes making up of human tandem repeats can detect these VNTRs loci, producing a different pattern of variable DNA fragments, so-called DNA finger prints. In or der to estimate the number of VNTRs loci with their allelic and linked pairs and to determine their mode of inheritance, this study was investigated the pattern of DNA fingerprints detected by pV47-2 multilocus probe for a Korean family consisting of both parents and 8 offsprings. Then, the following results were obtained. 1. The segregation of up to 31 heterozygous DNA fragments from both parents can be analyzed in a single sibship. 2. Among 16 heterozygous maternal-derived DNA fragments, one pair shows tight linkage, whereas two pairs out of 15 heterozygous DNA fragments derived from father is linked. 3. Two pairs out of 16 heterozygous DNA fragments derived from mother show allelism, whereas one pair of 15 heterozygous paternal-derived DNA fragments is allelic. 4. The number of different VNTRs loci detected by pV47-2 multi locus probe is 25 in the range of 4-30 Kb. Theoretically, 65 different VNTRs loci using pV47-2 multi locus probe could be detected if DNA fragments below 4 Kb in size were estimated. 5. It is possible to analyse up to 13 hypervariable VNTRs loci simultaneously in a single individual, a rate of genetic marker generation that is far highet than that obtained by conventional methods. 6. Heterozygous maternal DNA fragments are transmitted to 53.12% of the progeny, Whereas paternal fragments show 48.33% transmission. Then, the segregation ratio is consistent with 1:1. 7. The higher rate of genetic marker generation and the stable inheritance of variable DNA fingerprints detected by pV47-2 multi locus probe, together with the low population frequency of individual fragments, make them suited to use not only individual identification of forensic samples and parentage testing in the field of forensic medicine, but also to linkage analysis for genetic diseases.
